Metode yang digunakan penelitian ini adalah Rancangan Acak Lengkap (RAL). Yang terdiri 5 perlakuan yang berbeda yaitu : P1 = ( Tanah ) dengan perbandingan (1), P2 = (Tanah : Kompos : pasir ) dengan perbandingan (1:1:1), P3 = ( Tanah : Kompos ) dengan perbandingan (1:1), P4= ( Kompos : Pasir ) dengan perbandingan (1:1), P5 = ( Tanah : Pasir ) dengan perbandingan (1:1) Masing-masing perlakuan diulang sebanyak 3 kali dan setiap ulangan terdapat 5 sampel. Pada hasil analisis statistk diketahui bahwa tanaman dengan perlakuan P3 ( Tanah : Kompos ) dengan perbandingan (1:1) menunjukkan tinggi tanaman yang paling baik dengan ketinggian 1,6 cm dibandingkan dengan media tanam yang lainnya. Dan perlakuan P5 menunjukkan hasil tinggi tanaman yang paling rendah yaitu 1,1 cm. Hal ini di sebabkan oleh unsur hara yang terdapat pada perlakuan P3 memenuhi untuk pertumbuhan tumbuhan tanaman tembakau. Dari pengamatan menunjukkan bahwa pemberian kompos dapat menambah unsur hara pada media sehingga kebutuhan unsur hara pada media mencukupi untuk proses pertumbuhan batang tembakau
