diabetes mellitus (DM) is a disease characterized by elevated blood glucose levels (hyperglycemia) due to reduced insulin secretion , insulin or both. One indicator of diabetes, namely the destruction of pancreatic beta cells followed by increased expression of inducible Nitric Oxide Synthase (iNOS) in pancreatic beta cells. This study aims to determine the effect of freshly squeezed fruit squash on the expression of iNOS in the pancreatic beta cells in mice results Streptozotocin induction of Multiple Low Doze (MLD-STZ).
The research used experimental method to completely randomized design (CRD). 24 mice were 8 weeks old, weighing 20 ± 2 g divided into 6 treatments and 4 replications. Mice model of diabetes was obtained by inducting MLD-STZ intraperitoneally. DM mice is characterized by increased levels of fasting blood glucose> 126 mg / dl, then mice disonde with freshly squeezed fruit squash dose of 121 mg / 20 gBW, 242 mg / 20 gBW, 363 mg / 20 gBW, and 484 mg / 20 gBW. At the end of the treatment, mice were dissected to take organ pancreas. Pancreatic Organ preparations made and stained with immunohistochemical techniques using specific antibodies to see the expression of iNOS. The results showed that the juice of fruit squash have no effect on the expression of iNOS in pancreatic beta cells, but has a tendency to decrease the expression of iNOS in pancreatic beta cells.
