Nanopartikel dibuat dengan metode gelasi inonik caranya mencampurkan sitrat yang polianionik ke dalam kitosan yang polikationik setetes demi tetes menggunakan magnetic stirer. Hasil uji ukuran partikel diperoleh ukuran partikel formula I,II,III,dan IV sebesar 50,9nm, 92,1nm, 701,3nm, dan 143,3nm. Nanopartikel yang terbentuk lalu dilakukan uji penjeratan dan pelepasan zat aktif. Dari hasil uji penjeratan dan pelepasan zat aktif diperoleh bahwa nanopartikel formula I,II,III,IV persen penjeratan 59,42%, 81,56%, 45,81%, 64,40%. Nanopartikel formula II yang terbentuk diformulasikan menjadi sediaan gel yang akan diuji dengan evaluasi meliputi uji homogenitas, uji viskositas dan sifat alir, uji daya sebar, dan uji pH. Dari hasil uji evaluasi diperoleh bahwa gel nanopartikel stabil pada suhu kamar dengan organoleptik homogen, bau khas dan berwarna bening transparan, viskositas 245000-550000 cps yang diukur pada 3 rpm, kemampuan daya sebar 58,55-61,25 mm, pH 5,87. Hasil uji aktivitas antiacne terhadap sediaan gel mengandung 1% klindamisin nanopartikel memiliki daya hambat sebesar 29,5 mm yang dibandingkan dengan produk Medi-klin memiliki daya hambat sebesar 23,5 mm.
