Escherchia coli merupakan bakteri flora normal dalam tubuh manusia yang berasal dari kotoran hewan atau manusia, biasanya ditemukan pada hewan atau tanaman yang telah mati. Staphylococcus aureus merupakan flora normal kulit dan membran mukosa manusia lainnya menyebabkan supurasi, pembentukan abses, dan berbagai infeksi piogenik. Kaldu ayam merupakan suatu komposisi media mempunyai zat gizi yaitu karbohidrat, protein dan mineral yang dapat dimanfaatkan bakteri untuk pertumbuhannya.
Tujuan penelitian ini untuk mengetahui jumlah bakteri Escherchia coli dan Staphylococcus aureus pada media kaldu ayam dan media PCA metode ALT. Jenis penelitian ini deskriptif dengan sampel 1 ose bakteri Escherchia coli dan Staphylococcus aureus setelah pengenceran dan ditanam pada media kaldu ayam dan media PCA.
Hasil penelitian ini menunjukkan bahwa baik pada media kaldu ayam dan media PCA, bakteri Escherchia coli dan Staphylococcus aureus dapat tumbuh, namun terdapat perbedaan jumlah koloni bakteri. Dimana jumlah koloni pada media PCA lebih banyak dibandingkan dengan media Kaldu Ayam.
